By road: arriving from Toulouse on the N126, follow signs Lavaur, Graulhet and Puylaurens Centre, not Puylaurens, when you reach the roundabout that marks the start of the new bypass. When you enter the village, turn left at the traffic lights, then almost immediately turn right following the sign to Vielmur; go past the Ecole de Danse on your left, and count 17 plane trees again on your left, when you'll come to a small unmarked road to the left. Take this and go down the hill, swing left and you will see Pépoulie, the first house you come to on the right. It is set back about 75 metres from the road.
By air convenient airports are Toulouse and Carcassonne (one hour's drive away), Béziers and Perpignan (two hours' drive), and Montpellier and Girona/Barcelona (three hours' drive).
All of these are served by the budget airlines like Ryanair, easyJet, Brussels Airlines, OLT, Flybe, bmibaby and Germanwings.

Book your train journey to Toulouse-Matabiau central station, and you can get a local train to Vielmur-sur-Agout, 11 km from Pépoulie. There are trains every hour or so, the journey takes about an hour, and costs around EUR 12 per person per single journey.
Alternatively, fly to Toulouse-Blagnac airport, then take the Flybus shuttle to Toulouse coach station (gare routière), which is right next to Toulouse-Matabiau train central station. Buses depart every 20 minutes and the journey takes about 20 minutes depending on traffic. Then take the train to Vielmur-sur-Agout.
